Entering a political election as a candidate. The most common modern use of this word.
市長(しちょう)選挙(せんきょ)出馬(しゅつば)する。
To run for mayor.
彼女(かのじょ)来年(らいねん)国政(こくせい)選挙(せんきょ)への出馬(しゅつば)表明(ひょうめい)した。
She announced her candidacy for next year's national election.

2. entering a competition; stepping into the fray
More broadly, stepping forward to take on a challenge or competition. Originally referred to riding out to battle on horseback.
社長(しゃちょう)(みずか)出馬(しゅつば)して交渉(こうしょう)にあたった。
The president himself stepped in to handle the negotiations.
今回(こんかい)のプロジェクトには部長(ぶちょう)出馬(しゅつば)することになった。
The department head will be stepping in for this project.
ベテラン選手(せんしゅ)代表(だいひょう)(せん)出馬(しゅつば)する可能性(かのうせい)がある。
There is a possibility that the veteran player will enter the national team match.

Literally "riding out on horseback" ((しゅつ) = go out + () = horse). Originally a military term for riding into battle, now primarily used for political candidacy.
