To tell someone what to do in a work or organizational context. Implies a hierarchical relationship but is more neutral than 命令(めいれい)する.

上司(じょうし)が作業(さぎょう)内容(ないよう)を指示(しじ)する。

2. to indicate; to point out; to designate

To point to or identify something specific, such as indicating a direction or designating a particular item.

矢印(やじるし)が出口(でぐち)の方向(ほうこう)を指示(しじ)している。

The arrow indicates the direction of the exit.

地図(ちず)で場所(ばしょ)を指示(しじ)して説明(せつめい)した。

I explained by indicating the location on the map.

先生(せんせい)は黒板(こくばん)の図(ず)を指示(しじ)しながら説明(せつめい)した。

The teacher explained while pointing to the diagram on the blackboard.

In the workplace, 指示(しじ) is the standard neutral term for giving directions to subordinates — much softer than 命令(めいれい) (command). It's the everyday word for workplace instructions and is not perceived as harsh.
